Bestimmung der „C" vitamin im Harn : Bei der Bestimmung der Ascorbinsäure mit Methylenblau hat man mit zahlreichen Fehlerquellen zu rechnen; um diese zu vermeiden, ist es ° besonders wichtig, die PH jedes Mediums, in dem die Reaktion vor sich geht, genau einzuhalten. Auch bei der Einstellung der Versuchsbedingungen auf dieselbe PH hängt aber die Fähigkeit der Ascorbinsäure, Methylenblau zu reduzieren in hochem Masse vom Puffersystem ab. Da dieselbe Vitamin C Menge auch bei optimaler PH die gröste Methylenblau-Menge in Gegenwart von Acütat-Puffer reduzieren kann, wodurch der theoretische Faktor am ehesten erreicht wird, ist dieses System als das geeigneste anzusprechen. Diese Ansicht erscheint umsomehr berechtigt, .da das is der ursprünglichen Vorschrift von Bonsignore und Martini die Rolle des Katalysators spielende Natriumthiosulfat vollkommen überflüssig geworden ist, so dass man auch nicht mehr mit der Fehlerquelle zu rechnen hat, dass der im stark saueren Medium abgespalten Schwefel das Methylenblau weiter reduziere. Die zwischen Methylenblau und der Ascorbinsäure ablaufende Reaktion wird durch die Cupri- und Ferri-Ionen blos in grossem Grade gestört, da diese durch das entstehende Leukomethylenblau sofort zu Cupro- und Ferro-Ionen reduziert werden. Die reduktorische Wirkung von 0.1 mg Ascorbinsäure wird durch 0.1 bis 0.2 ccm der l°/o Cuprisulfatlösung scheinbar vollkommen gehemmt. Die zur Verwendung gelangende Methylenblaulösung und das Untersuchungsmaterial müssen daher vollkommen frei von Cupri- und Ferri-Ionen sein. Im -Harn gibt es zweifellos Stoffe (Urochrom B), durch die die reduzierende Wirkung der Ascorbinsäure gehemmt wird, bei entsprechender PH und bei Verwendung des Acetat-Puffers erreicht jedoch der durch die Hemmung entstandene Verlust bloss massige Grade. Der Verlust beträgt bei geringen Mengen (10 mg) höchstens 20—30°/o bei grösseren Ascorbinsäuremengen (50 mg) 6 bis 14"/'o. On the deiermination of vitamin C in Urine : The determination of ascorbic acid by means of the methylene blue method involves many sources of error. In Order to avoid these it is of great importance to maintain the ph of the medium in which the reaction takes place. Even at the same ph the ability of ascorbic acid to reduce methylene blue is primarily depending upon the buffer system Since even at an optimal ph the same amount of vitamin C is capable of reducing the greatest quantity of methylene blue in the presence of an acetate buffer this system is to be considered as the most adequate one. This consideration is the more justified as the use of sodiumthiosulfate, which is applied as a catalyzer in the original method af BonsignoreMartini, becomes entirely superfluous, and the source of error, caused by the fact that the sulphur released in highly acid medium may reduce methylene blue still further, is eliminated. The reaction between- methylene blue and ascorbic acid may be inhibited by even the smallest amounts of cupric and ferric ions as the developed ieukomethylene blue reduces them immediately to cupro and ferro ions. The reducing effect of 0.1 mg ascorbic acid may be completely inhibited by 0.1—0.2 cc. cf a. 1% solution of cupric sulphate. That is the reason why both the solution of methylene blue and the test material should be void of ferric and cupric ions. It is doubtless that urine contains substances (urochrome B) capable of inhibiting the reducing prower of ascorbic acid, nevertheless, the use of an adequate ph and acetate buffer diminishes the loss produced by the inhibition to but a moderate degree-. The loss duo to the inhibiting effect is, if most, 20—30% when small amounts of ascorbic acid (10 mg) are present while the presence of larger amounts (50 mg) diminishes the loss to but 6—14%.
