Hydrolysis is a fundamental sample preparation method for the determination of the composition of proteins. It makes difficult the determination of some amino acids, that they decompose during the hydrolysis with HC1 applied most frequently, so they can not be precisely determined with the other amino acids by amino acid analyser. In this paper the possibilities of the determination of cysteine are studied. After trying the methods, the one using oxidation with dimethylsulphoxide was found the most favourable and its optimal conditions were determined. The examinations were performed on standards and on natural materials. By the results data were obtained on the accuracy and reproducibility of the method. Zur Bestimmung der Aminosäurenzusammensetzung von Eiweiß ist ihre Hydrolyse eine generelle Vorbereitungsmethode. Die Bestimmung einiger Aminosäuren ist jedoch durch ihren Abbau während der am meisten angewandten Hydrolyse durch Salzsäure erschwert, so daß sie mit AAS nicht so exakt bestimmt werden können. Nach Erprobung mehrerer Methoden wurde die günstigste Methode: eine Oxydation mittels Dimethylsulfoxyds ausgewählt und die optimalen Anwendungs bedingungen ermittelt. Die Untersuchungen wurden mit Standards und mit natürlichen Materialien durchgeführt. Mit den Ergebnissen wurden Angaben über die Genauigkeit und Reproduzierbarkeit der Methode gesammelt.
