Die zur Bestimmung des Proteingehaltes entwickelte Methode gründet sich auf die selektive Bestimmung der Peptidkette. Das Wesen der Bestimmung besteht darin, dass das Protein in der Biuretreaktion eine stöchiometrische Menge Kupfers festbindet. Die Menge des nicht gebundenen Kupfers wird sodann an Sephadex G -2 5 Gel durch Absorption gebunden, und das an Protein gebundene Kupfer mittels Atomabsorptionsspektrophotometrie bestimmt. Diese Bestimmung wird durch Substanzen nichtproteiner Natur nur dann gestört, wenn das Reagens durch sie reduziert wird oder wenn sie m it dem Kupfer eine Komplexverbindung bilden. Die Methode ist zur Bestimmung der in den Weinen vorkommenden geringer Mengen von Proteinen geeignet, falls man vor der Bestimmung jene Substanzen vom Muster entfernt, die das Biuretreagens reduzieren können. The method developed for the determination of the protein content is based on the selective determination of the peptide chain. The determination consists essentially in the binding of copper by the protein in a stoichiometric amount during the biuret reaction. The copper amount which has not been bound is then bound on Sephadex G — 25 gel by absorption and the copper amount bound to protein determined by atomic absorption spectrophotometry. Substances of non-protein nature are interfering with this determination only in the case when they are reducing the biuret reagent or when they are forming a complex with copper. The method is suitable for the determination of the minute amounts of protein present in wines provided the substances reducing the biuret reagent are removed from the sample prior to the determination.
