Abstract

Die Verfasser arbeiteten die vollständige Trennung von 19 Aminosäuren auf einer Dünnschicht, einem Gemisch von Silicagel G und Cellulose MN 300 im Verhältnis 62 : 38 aus; Zur Entwicklung wurden in der ersten Dimension Isobutilalkohol 8 8%-ige Ameisensäure-Wasser 75 : 15 : 10, in der zweiten tertiäres Butanol-Methylaethylketon — 5%-ige Ammonialösung 50 : 30 : 20 als Laufmittel angewendet. 16 Aminosäuren im Konzentrationsbereich von 10 ~9 щпо1, getrennt auf der Dünnschicht, entwickelt mit — vermittels Cadmiunacetat stabilisiertem — Ninhydrin wurden a) Nach der Eluierung spektrophotometrisch durchschnittlich mit einer Genauigkeit von + 10%, b) vermittels direkter Densitometrie durchschnittlich mit einer Genauigkeit von + 2 0% bestimmt. The complete separation of 19 amino acids was worked out on a thin-layer composed of the 62 : 36 mixture of silicagel and cellulose MN 300. In the first dimension the chromatogram was developed with the mixture of isobutylalcohol — 8 8% formic acid — water (75 : 15 : 10), in the second dimension with terciary butanol — methyl ketone and 5% ammonia (50 : 30 : 20). 16 amino acids, falling in the 1 0 - 2 pM region of concentration, separated on thin-layer, and visualized by ninhydrin reagent, stabilized with cadmium acetate, were determined, after elution, with ± 10% accuracy by spectrophotometry. The same amino acids were determined by direct dosimetry, with +20% accuracy.
